SutroVax Announces Name Change to Vaxcyte

The new name more accurately reflects the Company’s mission to improve global health by developing superior and novel vaccines designed to prevent or treat some of the most common and deadly infectious diseases worldwide.

The new Vaxcyte name and logo connote the Company’s focus on developing vaccines to prevent or treat some of the most common and deadly infectious diseases worldwide, utilizing its exclusively-licensed cell-free protein synthesis platform and its proprietary know-how. As an example, Vaxcyte has leveraged this platform and know-how, utilizing site-specific conjugation, to develop its lead pneumococcal conjugate vaccine program to incite T-cell and B-cell-mediated (leukocyte) immune responses.

VAX-24, formerly referred to as SVX-24 and the lead vaccine candidate in Vaxcyte’s pipeline, is a preclinical, 24-valent pneumococcal conjugate vaccine (PCV) that includes 11 incremental strains over and above the 13 strains in Prevnar 13® and covers the 23 strains contained in Pneumovax 23®, which together make up today’s standard of care vaccination regimen for adults. Vaxcyte expects to submit an Investigational New Drug application for VAX-24 to the U.S. Food and Drug Administration in the second half of 2021 to evaluate it for the prevention of invasive pneumococcal disease (IPD).

Vaxcyte’s pipeline also includes VAX-XP, a PCV with an expanded breadth of coverage of at least 30 strains, including newly emerging strains responsible for IPD and antibiotic resistance, as well as a novel conjugate vaccine designed to prevent Group A Strep infections and a novel protein-based therapeutic vaccine designed to slow or stop the progression of periodontal disease by targeting the keystone pathogen responsible for this chronic, oral inflammatory disease.

In this role, Mr. Guggenhime will lead the finance, business and corporate development, strategy and communications activities at SutroVax.

Mr. Guggenhime brings more than two decades of leadership experience at both private and public healthcare companies, including as the chief financial officer of biotechnology companies Dermira, Calistoga Pharmaceuticals and Facet Biotech among others.

Mr. Guggenhime joins SutroVax from Dermira, Inc., where he served as Chief Financial Officer from April 2014 through the acquisition of the company by Eli Lilly and Company in February 2020. During his time at Dermira, Mr. Guggenhime executed on a wide range of financial and strategic transactions, including the company’s initial public offering, follow-on offerings and the sale of the company; planned and executed the transition to a commercial-stage organization in connection the company’s first product launch; and built and led business planning and process initiatives for multiple corporate functions. Prior to Dermira, Mr. Guggenhime served as Chief Financial Officer for CardioDx, Inc., a privately held commercial-stage cardiovascular molecular diagnostics company. He also served as Chief Financial Officer for Calistoga Pharmaceuticals, Inc., a privately held, development-stage company, which was acquired by Gilead Sciences, Inc. in 2011; and as Chief Financial Officer for Facet Biotech Corporation, a biotechnology company that was acquired by Abbott Laboratories in April 2010. He previously served as Chief Financial Officer of PDL BioPharma, Inc. until Facet Biotech was spun off from PDL BioPharma in December 2008. Prior to joining Facet Biotech, Mr. Guggenhime served as Chief Financial Officer for Neoforma, Inc. a provider of supply chain management solutions for the healthcare industry, which was acquired by Global Healthcare Exchange, LLC in March 2006. SutroVax’s lead vaccine candidate, SVX-24, is a preclinical, 24-valent pneumococcal conjugate vaccine (PCV) that includes 11 incremental strains over and above the 13 in today’s standard of care, Prevnar 13®, including strains responsible for high case-fatality rates, antibiotic resistance and/or meningitis. Preclinical proof of concept studies for SVX-24 evaluating the surrogate immune endpoints used for adults and children demonstrated comparable responses to the 13 common strains in Prevnar 13 and superior responses to the 23 common strains in Pneumovax®23, the non-conjugate polysaccharide-only vaccine. The pneumococcal vaccine class is the industry’s largest, accounting for over $7 billion in annual sales. These SVX-24 preclinical data have been accepted for presentation at the 12th International Symposium on Pneumococci and Pneumococcal Disease (ISPPD-12) anticipated to take place in June 2020.

The SutroVax PCV pipeline also includes SVX-XP, a PCV with an expanded breadth of coverage of at least 30 strains, including newly emerging strains responsible for invasive pneumococcal disease and antibiotic resistance. SutroVax has generated preclinical proof of concept versus the standard of care vaccines and intends to advance SVX-XP into the clinic as a future successor to SVX-24.

About Pneumococcal Disease and the Pneumococcal Vaccine Market

Pneumococcal disease is an infection caused by Streptococcus pneumoniae. This infection can lead to a wide range of serious primary infections including pneumonia, meningitis and blood stream infections, as well as ear and sinus infections, and can also cause secondary infections due to other respiratory pathogens. According to the Centers for Disease Control and Prevention (CDC), an estimated 900,000 people suffer from pneumococcal pneumonia in the United States each year, including as many as 400,000 requiring hospitalizations and approximately 28,000 deaths.  The market-leading vaccine is a 13-valent PCV, Prevnar 13, that has worldwide annual sales of approximately $6 billion, yet does not protect against a significant number of circulating strains of pneumococcus causing invasive disease in adults and children.

SVX-24 is designed to improve upon the 13-valent PCV standard of care by covering the additional eleven strains that are responsible for the majority of residual pneumococcal disease currently in circulation in both the United States and European Union and associated with high case-fatality rates, antibiotic resistance and/or meningitis. The company believes it can achieve this by employing its cell-free protein synthesis platform, which is comprised of the XpressCFTM platform exclusively licensed from Sutro Biopharma and its proprietary know-how that offers several advantages over conventional cell-based protein expression methods, which the company believes enables it to generate a more broad-spectrum PCV. SutroVax believes it can add more antigenic strains without compromising the overall immune response. In particular, SutroVax’s ability to specify the attachment point of antigens, including polysaccharides, on protein carriers represents a significant improvement over the random conjugation that occurs with conventional PCVs. This site-specific conjugation is designed to ensure that T-cell epitopes on the protein carrier are optimally exposed, maximizing the immune response, whereas random conjugation can block these critical immunogenic epitopes, dampening the immune response and causing a phenomenon known as carrier suppression. The company believes this precise control of conjugation chemistry enables it to design broader-spectrum conjugate vaccine candidates using carrier-sparing conjugates that use less protein carrier without sacrificing immunogenicity.

SutroVax achieved preclinical proof of concept for SVX-24 by demonstrating that SVX-24 has the potential to protect against the pneumococcal strains collectively covered by Prevnar 13 and Pneumovax 23 and showing the boostable immune responses that are the hallmark of a conjugate vaccine. SVX-24 includes 24 purified capsular polysaccharides of Streptococcus pneumoniae (1, 2, 3, 4, 5, 6A, 6B, 7F, 8, 9N, 9V, 10A, 11A, 12F, 14, 15B, 17F, 18C, 19A, 19F, 20, 22F, 23F and 33F), each of which is conjugated to eCRM, the company’s proprietary protein carrier, to produce 24 monovalent conjugates. These conjugates are mixed into a final vaccine formulation and then adsorbed to alum.

There are critical differences between SVX-24 and other currently available PCVs relating to SutroVax’s protein carrier, eCRM, particularly the ability to perform site-specific conjugation and use milder reaction conditions for conjugation. SutroVax achieves site-specific conjugation through the insertion of multiple non-native amino acids (nnAAs) into the protein carrier, which is not possible with the conventional chemistry used for making other PCVs. The click chemistry SutroVax uses for site-specific conjugation of the polysaccharides to the nnAAs on the protein carrier may also minimize damage to the critical immunogenic epitopes on both the protein carrier and the polysaccharides through milder and shorter reactions, while other PCVs use conventional chemistries that involve harsher and longer reaction conditions. SVX-24, as tested in preclinical studies, showed nearly double the serotype spectrum of coverage of Prevnar 13, yet contains a similar amount of protein carrier. The company believes the resulting decreased carrier burden per conjugate of SVX-24 is critical for avoiding carrier suppression and producing broader-spectrum PCVs without sacrificing immunogenicity.

Jim Wassil appointed Chief Operating Officer

In this role, Mr. Wassil will lead clinical development, regulatory affairs, medical affairs, quality assurance and program management activities.

Mr. Wassil brings extensive experience in the development and commercialization of vaccines having spent the past three decades in positions of increasing responsibility in the vaccine divisions of Merck, Novartis, and Pfizer. Most recently, he was responsible for market access, policy, pricing, tender negotiations, epidemiology and health outcomes as the business unit lead for Pfizer Vaccines, where he was responsible for the full vaccine portfolio, including both the pneumococcal conjugate vaccine franchise (Prevnar 13) and the meningococcal conjugate vaccine franchise (Nimenrix, Neisvac-C and Trumemba). SutroVax Receives CARB-X Award for Up to $15 Million to Develop a Universal Vaccine to Prevent Group A Streptococcus Infections

Proprietary conserved antigen with potential to provide broad protection against strains of Group A Strep licensed from University of California San Diego

Foster City, CA, September 3, 2019SutroVax, Inc., a biopharmaceutical company dedicated to the development of best-in-class conjugate vaccines and novel complex antigen-based vaccines to prevent serious infectious diseases, today announced it has received an award from CARB-X for up to $15 million in non-dilutive funding to develop a universal vaccine to prevent infections caused by Group A Strep bacteria, which include pharyngitis, impetigo, and necrotizing fasciitis. The award commits initial funding of up to $1.6 million and up to $15 million in total funding available upon achievement of development milestones.

“This funding will support our collaborative efforts to develop a safe, effective Group A Strep conjugate vaccine where none exist today to prevent a highly prevalent disease with widespread morbidity and mortality,” said Jeff Fairman, Ph.D., Vice President Research and co-founder of SutroVax. Innovative Group A Streptococcus Vaccine

SutroVax’s Group A Strep vaccine is based on a discovery at the University of California, San Diego, that led to the development of a conserved cell wall polysaccharide antigen with the potential to provide broad protection against strains of the bacteria, while avoiding cross-reactive antibodies that could target human tissue. SutroVax exclusively licensed the rights to this patented antigen and is developing the Group A Strep vaccine, by utilizing its proprietary conjugation technology, which permits site-specific conjugation of polysaccharide antigens to immunogenic proteins and preservation of critical B-cell and T-cell epitopes on the carrier protein, resulting in improved conjugate vaccines. Conjugate vaccines have led to multiple highly effective vaccines to prevent disease caused by pneumococcus, meningococcus, and Haemophilus influenzae B bacteria and represent the largest segment of the global vaccines market.

About Group A Streptococcus

Group A Streptococcus is a serious human pathogen causing an estimated 600 million cases of pharyngitis annually worldwide and increasing cases of severe invasive infections, including sepsis, necrotizing fasciitis, and toxic shock syndrome.1 Pharyngitis is highly prevalent in school-age children and a significant driver of antibiotic prescriptions worldwide. The growing use of antibiotics contributes to antibiotic resistance, which can threaten the ability to prevent the spread of infections and lead to higher medical costs, prolonged hospital stays, and increased mortality. In the United States, the Centers for Disease Control and Prevention (CDC) estimates that 23,000 people die each year from drug-resistant bacterial infections. In Europe, the European Centre for Disease Prevention and Control (ECDC) estimates that there are 33,000 deaths annually from the rise of drug-resistant bacteria. Group A Strep is also responsible for post-infectious immune-mediated rheumatic heart disease (RHD), a leading cause of mortality in the developing world. An estimated 30 million people are currently affected by RHD, with over 500,000 deaths annually (60% under the age of 70) and over 11 million disability-adjusted life years lost.2 Despite high global demand, there is currently no commercially available vaccine against Group A Strep.

Research details preclinical use of vaccine targeting keystone pathogen Porphyromonas gingivalis

Foster City, CA, February 25, 2019SutroVax Inc., a biopharmaceutical company dedicated to the delivery of best-in-class conjugate vaccines and novel complex antigen-based vaccines to prevent serious infectious diseases, today announced the publication of research highlighting the importance and effectiveness of a proprietary multi-valent vaccine that has demonstrated protective immunity against Porphyromonas gingivalis (Pg) – a common bacteria believed to be responsible for periodontal disease.

The published research describes the use of three recombinant proteins from P. gingivalis, namely minor fimbriae protein (Mfa1), RgpA gingipain hemagglutinin domain 1 (HA1), and RgpA gingipain hemagglutinin domain 2 (HA2).  This vaccine combination was designed to interrupt biofilm formation between P. gingivalis and other oral bacteria which stimulates virulence factors that result in disease progression and circumvent the host immune response.  The published results demonstrate that immunization with the combination vaccine induces high levels of Pg protein specific Immunoglobulin G (IgG) antibodies, that resulted in significant protection against bone loss elicited from P. gingivalis infection in a murine challenge model.

“The treatment and prevention of disease progression caused by P. gingivalis has the potential to significantly impact dental health.  P. gingivalis has also been shown to be associated with increased risk for heart attack, stroke, and most recently Alzheimer’s Disease,” said Jeffery Fairman, Ph.D., SutroVax, Inc.’s vice president of research, co-founder and author on the publication. “These findings provide compelling preclinical evidence that a vaccine targeting the biofilm cascade may be an efficacious approach for the treatment and/or prevention of periodontal disease.”

The paper, published in the Journal of Clinical Periodontology and authored by researchers at the University of Florida College of Dentistry, Boston University School of Medicine, Section of Infectious Diseases and SutroVax, describes the application of SutroVax’s proprietary Xpress CFTM cell free protein synthesis (CFPS) technology as a platform to produce vaccines targeting Pg infection.

“Periodontal disease affects over 500 million people worldwide and over half of American adults over 40 years of age are suffering from measurable oral bone loss from the disease” said Grant Pickering, CEO and co-founder. “The application of our cell free technology offers a promising avenue to intercept periodontal disease and its resultant morbidity.”

“These results support that a vaccination approach may provide an opportunity to change the trajectory of periodontal disease,” said Frank C. SutroVax Announces $85M Series C Financing led by TPG Growth

Other new investors include Medicxi and Foresite Capital.  All of SutroVax’s existing institutional investors, including Abingworth, Longitude Capital, Frazier Health Care Partners, Pivotal bioVenture Partners, Roche Venture Fund, and CTI Life Sciences Fund also participated in the financing.

Proceeds of the financing will be used to advance the company’s potentially best-in-class broad-spectrum pneumococcal conjugate vaccine (PCV) candidate through mid-stage clinical development and continue to expand its pipeline of novel vaccines.  SutroVax’s PCV is designed to prevent invasive pneumococcal disease caused by Streptococcus pneumoniae and is uniquely positioned to significantly expand protection against circulating strains of pneumococcus.

SutroVax is also announcing a strategic contractual relationship with Lonza Pharma & Biotech, one of the world’s leading contract manufacturers of complex biologics, to manufacture its PCV program via an end-to-end supply relationship. The parties have established strong alignment on process and conjugation development, as well as microbial manufacturing scale-up for drug substance at Lonza’s Visp, Switzerland, site. This close technical collaboration between SutroVax and Lonza will support the successful delivery of this potentially best-in-class vaccine to the clinic as efficiently as possible and to streamline its advance to the market.

Pneumococcal disease is an infection caused by Streptococcus pneumoniae. This infection can cause a wide range of serious illnesses including pneumonia, meningitis and blood stream infection as well as ear and sinus infections. In the U.S., the CDC’s Advisory Committee on Immunization Practices (ACIP) recommends all children aged two months to five years and immunocompromised children aged six years and older be vaccinated with Prevnar 13®.  In addition, the ACIP recommends all adults aged 65 years and older and immunocompromised adults aged 19 years and older be vaccinated with Prevnar 13®and Pneumovax®, a 23-valent non-conjugate vaccine with over $600 million in annual sales.
